Topical PK-PD Dose-Response and Formulation Optimization of Melatonin for AD

Overview

Several studies have investigated the influence of Topical Melatonin in various skin conditions. Specific studies show that it has promise in chronic Atopic Dermatitis (AD) treatment, due to the soothing properties it has. In the past, I used MathWorks MATLAB Simbiology Quantitative Systems Pharmacology (QSP) in order to see the effect of topical melatonin on Atopic Dermatitis. Focusing specifically on modeling the pathology of AD. I found that it showed promise for being a potential treatment. Due to its anti-inflammatory properties In this study, I directly investigated how topical melatonin works within the skin. Using Pharmacokinetics and Pharmacodynamics principles. Working directly off of my previous model, I found that topical melatonin was effective for AD. It reduces inflammatory biomarkers, while moderately raising antioxidantal biomarkers. Thus, allowing AD-induced skin to have similar skin homeostasis to regular skin. These positive effects were beneficial up to a certain point. Above 5mM per dose benefits plateau. Therefore raising the risk of antibiotic resistance.
