The Effects of Basil and Oregano on Xanthomonas Vasicola Pathovar Vasculorum

Overview

The question tested in this experiment is “How will the essential oils of basil and oregano affect the growth of Xanthomonas vasicola pathovar vasculorum?”. Bacterial Leaf Streak is caused by Xanthomonas vasicola pv. Vasculorum is a newly diagnosed pathogen affecting the foliar of corn. It has been hypothesized that both basil and oregano will be effective antimicrobial agents. Research taken from the medical and agricultural fields have shown basil and oregano essential oils are effective against the growth of various pathogenic bacteria. Fresh basil and oregano were pulverized and bathed in distilled water. After juicing the herbs, the herbal water solution was run through a separatory funnel using dichloromethane to extract the essential oils from the water. This solution was then run through a Rotary Evaporator, to evaporate the dichloromethane from the essential oils. Dimethylformamide was mixed with the essential oils to create an experimental solvent. Two tests were then used to test the antimicrobial effects of basil and oregano. First, the Kirby Bauer disk diffusion test was performed by placing six filter disks on painted plates of Xanthomonas vasicola pv. Vasculorum. The essential oils were placed on filter disks, and incubated and 30 degrees Celsius for approximately 10 hours. This test was also preformed using E. coil for a negative control. The second test preformed was serial dilution test. The initial results show that the essential oils of basil and oregano did inhibit the growth of Xanthomonas vasicola pv. Vasculorum. Statistics were used to analyze the data.
