Investigation of the Therapeutic Potential of Umbilical Cord Tissue Derived Mesenchymal Stem Cells

Overview

Introduction: Inflammation and infection are important in the etiopathogenesis of pulmonary diseases. Umbilical cords are usually discarded postdelivery. Mesenchymal stem cells (hMSCs) derived from cord tissue were evaluated for their functional potential to treat infection and inflammation. Methods: hMSCs were cultured with or without lipopolysaccharide (LPS) or peptidoglycan (PEP). hMSC supernatants were analyzed for secreted cytokines using Luminex and for gene expression utilizing RT-PCR. hMSCs antimicrobial activity was measured by growth of Staphylococcus aureus (SA) and Pseudomonas aeruginosa (PA) and functionality by anti-inflammatory activity on epithelial cells. Results: hMSCs secreted cytokines IL-6 and IL-8. LPS and PEP enhanced cytokine secretion. hMSCs expressed genes for IL-6, IL-8, and MIP-3α. LPS increased expression of IL-6 and IL-8, while PEP decreased IL-6 and MIP-3α. hMSC supernatants were antimicrobial; they decreased SA CFUs from 150 to 68.5 and PA CFUs from 208.4 to 35.3 respectively after treatment for 2 hours (n=3, p≤0.05). Cord tissue derived hMSCs supernatants had anti-inflammatory activity, which was enhanced by LPS (5.64 vs 6.56 cycle difference). Conclusions: hMSCs secrete a variety of cytokines and display antimicrobial and anti-inflammatory activity. Donor variability was demonstrated. Donor related variability in the phenotypic properties might be important in the functionality of hMSCs.
