Investigating the proliferation, invasion, and resistance to chemotherapy-induced apoptosis of A549 lung cancer cells in relation to synthetic nicotine exposure

Overview
Marketed as the tobacco-free alternative to traditional rolled cigarettes, e-cigarettes use synthetic nicotine to bypass FDA standards for tobacco products. Synthetic nicotine (TFN), a racemic mixture of R and S nicotine in equal proportions, has been understudied in regard to its metabolic and pharmacological effects due to the relatively recent advancements in TFN use. The purpose of this research is to investigate the behavior of A549 cells to determine the proliferation, invasion, and resistance to chemotherapy of lung cancer cells in relation to TFN exposure. (S)-(-)-nicotine acted as a positive control throughout the research due to the extensive research of tobacco-derived nicotine on A549 cell behavior, while unstimulated cells acted as a negative control. The samples treated with TFN demonstrated a greater mean proliferation by 4.28%, 5.98% lower than the positive control of S-(-)-nicotine. The TFN sample displayed decreased average caspase-3 activity of 8.73%, increased ATP production by 32.03%, and increased average ratio of red to green fluorescence in the mitochondrial membrane by 21.17%. Invasion and metastasis was measured through invasion assays and wound healing assays, with the TFN demonstrating an increased invasion rate of 29.7% and migration of 20.83%. When exposed to TFN, the cells demonstrated the hallmarks of a more severe lung cancer, with higher proliferation and invasion rates as well as increased resistance to chemotherapy-induced apoptosis. As a result, this research can shed light on the potential negative health effects of e-cigarettes, particularly in relation to lung cancer.
