Up-Regulating the Gaba and Pkg Pathways as a Potential Therapy for Epilepsy

Overview

In many prokaryotes, the genetic locus termed clustered regularly interspaced short palindromic repeats (CRISPR) and the CRISPR-associated (Cas) genes encode RNA-guided nucleic acid recognition systems that confer adaptive immunity against invading viruses for many bacteria and archaea. The Cas protein Csm6 contributes to CRISPR immunity by targeting host transcripts in order to induce dormancy or promote programmed cell death of the host. Here we use purified Csm6 from Lactococcus lactis (Llcsm6) and in vitro cleavage analysis to show that Csm6 proteins cut RNA using its HEPN domain. Together, these results demonstrate the importance of relating the structure of Csm6 to its function to utilize the protein in genetic editing, such as that commonly seen with CRISPR/Cas9.
