Expression and Characterization of Tardigrade CAHS Protein Q
Overview
The tardigrade protein CAHS D has been shown to stabilize proteins such as lactate dehydrogenase at high temperatures. This experiment focused on the stabilization abilities of another CAHS isoform, CAHS Q, on alcohol dehydrogenase, another oxidoreductase enzyme. Alcohol dehydrogenase is used to convert ethanol into acetaldehyde, and vice versa. CAHS Q’s expression was optimized to be 250 µM final IPTG incubated at 16°C for 16 hours. This was confirmed by SDS-PAGE analysis. This condition was then used for the protein’s FPLC purification in which the positive CAHS Q protein was subjected to anion exchange chromatography. The procedure purified the protein, which was then dialyzed and lyophilized.
