Exosome-Packed Phenyl Boronate, a Redox Scavenger as an Effective Sensitizer for Immunotherapy in Cervical Cancer

Overview

Cervical cancer is the 2nd largest among all gynecological cancer cases reported worldwide and is a challenge to treat. Immunotherapy remains a widely recognized treatment option but suffers from challenges such as T cell exhaustion. Redox intermediates released by tumor cells were reported to limit the effectiveness of T cell therapy. Based on the above, I devised a bioengineering strategy to construct a T cell prototype involving packing of a peroxynitrite inhibitor in endogenous exosome vehicles and loading them into CD8+ve T cells. My results showed that the PBA-packed exosome construct inserted into CD8+ve T cells decreased peroxynitrite release from cancer cells and induced apoptosis as determined by staining if Annexin-V and Propidium Iodide, thus leading to effective cancer cell killing. My results have far-reaching implications in cancer immunotherapy as they provide a new paradigm to overcome the limitations of immunotherapy that involve T cells.
