Characterization of the ELOF1-CE022 Protein Complex Awan Awan Lynn Vocational Technical Institute 12th Grade The primary objective of this research is to determine if ELOF1, a transcription elongation factor, directly interacts with CE022, an understudied protein. Based on AlphaFold screenings and proteomic data, we hypothesize that CE022 associates with ELOF1 and prevents it from associating with RNA polymerase II. To test whether CE022 and ELOF1 interact, I cloned and performed test expression and protein purifications to obtain CE022. I then analyzed whether ELOF1 and CE022 interact using a pulldown assay and analytical size exclusion chromatography. These experiments demonstrated that ELOF1 and CE022 directly interact. Finally, I compared the ELOF1-CE022 AlphaFold3 prediction with known RNA polymerase II-ELOF1 structures. This analysis showed that the ELOF1-CE022 complex would not be expected to bind RNA polymerase II. These findings, supported by computational predictions, suggest that CE022 confines ELOF1 from binding RNA polymerase II thereby establishing CE022 as a potential regulator of ELOF1.
