Blue-reka! Investigating Methylene Blue as a Neuroprotectant for Traumatic Brain Injury Using a Novel Drosophila Model

Overview

Traumatic brain injury (TBI) is the leading cause of disabilities and deaths worldwide, with Americans experiencing 2.5 million hospitalizations and fatalities annually and $19B in economic losses. TBI’s physical and mental impacts decrease quality of life and increase the risk of future neurodegenerative conditions. This project investigates methylene blue’s effect on mitigating secondary injury after TBI, using Drosophila hydei as a novel in vivo treatment model. This research will increase knowledge for medical sciences about well-tolerated, affordable, and accessible neuroprotection to mitigate TBI. HYPOTHESIS: IF drosophila consume methylene blue pre- and post- traumatic brain injury, THEN it will mitigate secondary brain injury, with the highest dosage being most effective, BECAUSE methylene blue is neuroprotective, thus decreasing oxidative stress and regulating cell processes. PROCEDURE: 350 drosophila over 7 trials consumed various methylene blue dosages for 5 days, then received 3 strikes from a 90-degree retraction angle in the High-Impact Trauma (HIT) device. A 20-second climbing assay measured the number of drosophila passing a 2.5cm-high “finish line.” After 24 hours, survival counts were taken. OBSERVATIONS/DATA/RESULTS: The highest number of drosophila passing the finish line and surviving after 24 hours consumed 8 mg/kg of methylene blue. All treatment groups had higher average numbers of drosophila passing the line and surviving vs. Control. The Climbing-Survival Index (CSI) score – a novel biomarker I propose, combining immediate and delayed measures – was highest for the 8 mg/kg dosage. CONCLUSIONS: Methylene blue is neuroprotective, decreasing oxidative stress and regulating cellular processes to mitigate secondary injury. This research impacts healthcare and medical sciences, identifying a well-tolerated, affordable, and accessible neuroprotectant to improve TBI outcomes.
